This antibody is recommended for detection of human MKLP1 only. AKIN06 is not recommended for use with extracts from other species.

Material
AKIN06 is provided as an affinity purified rabbit polyclonal antibody. The antibody was raised against the motor domain of human kinesin MKLP1 (KNSL5, KIF23). Native MKLP1, with a predicted molecular weight of 98 kDa, is detected on Western blots of mitotic HeLa cell extracts. CH01, an MKLP1 isoform produced by alternative splicing, is detected in some cell extracts at 110 kDa. A nonspecific band at approximately 200 kDa may also be present (Fig 1). A mitotic HeLa cell extract is included as a positive control for antibody reactivity (see Fig. 1 and western blot instructions on p. 2). AKIN06 (50 µg of protein) is supplied as a lyophilized white powder.

Other lanes shown on the blot are recombinant motor domains of CENPE (lane 2), Chromokinesin (lane 3), Eg5 (lane 4), KIF3C (lane 5), MCAK (lane 6), Kinesin Heavy Chain (lane 7). For all motor proteins besides MKLP1, 100 ng of recombinant protein was used. MKLP1 was the only motor protein that was detected.

Immunofluorescence staining of MKLP1 (red) in mitotic HeLa cells is shown using 500 ng/ml (1:500 dilution) of AKIN06 antibody. Primary antibody was detected with a 1:500 dilution of goat anti-rabbit rhodamine conjugated antibody (Cat. # RG05). DNA (blue) was stained with 100 nM DAPI in PBS. Localization of MKLP1 within mitotic cells to the spindle midzone region and the midbody is in agreement with published data (Matuliene and Kuriyama, 2002. Mol. Biol. Cell 13:1832-1845). The photograph was taken with a 100X objective lens. |
