Actin polymerization conditions

What procedure do you recommend for actin polymerization ?

Answer:
We recommend the following procedure for polymerizing actin:

  1. Dilute actin to 0.4 mg/ml with A-buffer (Cat. # BSA01 plus ATP: 1 x = 5 mM Tris-HCl pH 8.0, 0.2 mM CaCl2, 0.2 mM ATP) mix and leave on ice for 1 h.
  2. Add polymerization inducer (Cat. #. BSA02: 10 x = 500 mM KCl, 20 mM MgCl2 and 10 mM ATP) mix and incubate at room temperature for 1h.

For more critical work, cold spin at 150,000 x g for 2 h after step 1 to remove small oligomers of actin.
