Introduction

RhoA is one of most extensively studied member of Rho GTPase family. This family is involved in a wide range of cellular responses, including cytoskeletal reorganization, regulation of transcription, cell migration, cellular transformation and metastasis. RhoA is the predominant Rho isoform that is expressed in a large number of different cell types.

This antibody is recommended for detection of RhoA in human, mouse, rat or other extracts. The following protocols have been tested with this anti body: 

  • Western blot analysis:  recommended 
  • Immunocytochemistry:  recommended

Material

The anti-RhoA antibody (Cat. # ARH04) is a mouse monoclonal antibody that only recognizes RhoA, not , RhoB, RhoC, Rac1, Rac2, Rac3, Cdc42 or H-Ras. Other commercially available RhoA antibodies recognize one or more of these close isoforms  Platelet extract (Cat. # EXT01) is included as a positive control. ARH04 is supplied as a lyophilized white powder.

Figure 1: Western blot analysis of purified small G-proteins using ARH03 as the primary antibody.. Recombinant small G-proteins and platelet extract were separated by SDS-PAGE and transferred to PVDF membrane.  Anti-RhoA antibody was diluted to 500 ng/ml (1:500) in PBST and the signal developed with a sensitive chemiluminescence reagent. Lane 1; 25 ng RhoA-6xHIS, Lane 2; 250 ng RhoB-6xHIS, Lane 3; 250 ng RhoC-6xHIS, Lane 4; 250 ng Rac1-6xHIS, Lane 5; 250 ng Cdc42-6xHIS, Lane 6; 40 µg human platelet extract. Note: RhoA-6xHIS runs at 28kDal whereas the native RhoA in platelet extract runs at 23kDal.

Figure 2: Western blot analysis of cell extracts using ARH04 as the primary antibody.  Chemiluminescence detection of  RhoA in cell extracts (50 µg each) of rat NRK cells (lane 1), human HeLa cells (lane 2), bovine brain extract (lane 3), and human platelet cell extract (lane 4). The RhoA band is located at approx. 22kDal.  The blot was probed with a 500 ng/ml (1:500) dilution of anti-RhoA antibody.  Molecular weight markers are from Invitrogen.

Question 1:  What is the antigen that this antibody was raised against?

Answer 1:  The RhoA specific antibody (Cat. # ARH04) is a mouse monoclonal antibody and it was raised against amino acids 120-150 of the human RhoA protein.

 

Question 2:  What species has this antibody been tested against?
