Support in precision oncology

Objective

A Fortune 500 biopharmaceutical company required a comprehensive off-target counter-screen to evaluate the specificity of anti-kinesin drug candidates targeting KIF18A.

Result

Four compounds plus control inhibitors were tested in a motor panel consisting of KIF2, KIF3C, KIFC3, KIF4A, KIF10(CENPE), KIF11(Eg5), KIF20A, and KIF23. IC50s confirmed that the compounds were highly specific to KIF18A and its close homologs KIF18B and KIF19A.

Some of the data generated by Cyto was included in a reported by Amgen Research company (CA, USA) and Broad Institute (MA, USA) in 2024 (Payton M. et al. Small-molecule inhibition of kinesin KIF18A reveals a mitotic vulnerability enriched in chromosomally unstable cancers. Nature Cancer. 5, 66-84 (2024): Extended data Fig 2).
