Optimizing SAR for FtsZ

The objective

Our client submitted a focused library of small-molecule compounds for screening against the Staphylococcus aureus FtsZ protein, with the objective of identifying potential inhibitors to support antimicrobial drug development efforts.

The result

Cytoskeleton, Inc. optimized two complementary assay formats to evaluate modulation of FtsZ activity. The first assay measured polymerization via light scattering, which reflects the formation of higher-order filamentous structures. The second assay was a GTPase activity assay, which quantified the intrinsic enzymatic function of FtsZ. Following an initial screen of the parent compound library, successive rounds of structure–activity relationship (SAR) optimization were conducted, ultimately yielding sub-micromolar inhibitors with improved potency.
