• Buffer of choice for tubulin proteins

Supplied as a lyophilized powder. When resuspended in deionized water, it provides a 1x working solution: 80 mM PIPES pH 7.0, 2 mM magnesium chloride, 0.5 mM ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA).

Storage and handling

• After reconstitution, the buffer is stable and stored at 4°C
• Supplementation with 1mM guanosine triphosphate (GTP) BST06 to create an optimal tubulin buffer. As GTP is labile and will hydrolyze, GTP should be added fresh just before use. The GTP-supplemented buffer should not be stored.
• Glycerol is often added to a final concentration of 5-10% to enhance tubulin polymerization BST05. Glycerol is not necessary for the maintenance of biologically active tubulin.

All reagents used in this buffer are ≥ American Chemical Society (ACS) grade and ≥99% pure
