• Use as an affinity reagent for enrichment of active (GTP-bound) Rac and Cdc42 proteins

The Rac/Cdc42 binding domain of the human p21-activated kinase1 protein (PAK) has been overexpressed as a GST-tagged recombinant protein in a bacterial expression system.

Protein purity is assessed by scanning densitometry of Coomassie Blue-stained protein on a 4-20% polyacrylamide gel. Purity was determined to be ≥88% pure.

The biological activity of PAK01 is assessed by its ability to selectively bind the active forms of Rac1 and Cdc42 proteins in GTPγS-loaded lysates, compared to the inactive forms in GDP-loaded lysates (see datasheet). In this assay, the GTPγS-loaded samples typically show a 7–15-fold increase in Rac1 and Cdc42 signal compared to the GDP-loaded samples.
