Anti-SUMO-2/3 antibody (ASM23) was raised against full-length recombinant SUMO-2 protein (Uniprot: P61956) combined with a proprietary mix of peptides that include CQIRFRFDGQPINE. Epitope mapping has identified that the antibody recognizes a sequence/structure within the peptide CQIRFRFDGQPINE. It detects a wide range of SUMO-2/3-targeted proteins. ASM23 has been conjugated through its amine groups to the carbohydrate groups of horseradish peroxidase (HRP) to make ASM23-HRP. The antibody is purified via Protein G chromatography.

Reactivity:

• Broad cross-species reactivity, the peptide epitope sequence is conserved in mammals, birds, and amphibians.

• ASM23-HRP-S: 1 x 25 µl when reconstituted (37.5-50 µg per tube)

Formulation

• Buffer composition after reconstitution in 50% glycerol in water: 200 mM PIPES pH7.0, 1% sucrose, 1% dextran, 50% glycerol, 10 mg/ml BSA

Sensitivity:

• Detects ≤0.2 ng rSUMO-2 protein

Specificity:

• Highly selective: Detects ≤0.2 ng rSUMO-2 protein, will not detect 1000 ng of rSUMO1—demonstrating a ≥5000-fold specificity.
• In 3T3 cell lysates, high molecular weight SUMO2/3-modified proteins are abundant. SUMO2/3 signal decreases by over 60% with SUMO-2 shRNA knockdown and increases more than 10-fold after heat shock at 42 °C.
